#Interleukin2 #Immunotherapy reveals subsets of human #Tregs with distinct functional & tissue-homing characteristics | A "Shot-in-the-Arm" for #SLE 👍 | Open Access Study by Onur Boyman & Co. breaking at #Immunity Cell Press | Due to its stimulatory potential for immunomodulatory CD4+regulatory T (Treg) cells, low-dose interleukin-2 (IL-2) immunotherapy has gained considerable attention for the treatment of autoimmune diseases. In this investigator-initiated single-arm non-placebo-controlled phase-2 clinical trial of low-dose IL-2 immunotherapy in systemic lupus erythematosus (SLE) patients*, Miro E. Raeber, MD, PhD, et al generated a comprehensive atlas of in vivo human immune responses to low-dose IL-2. They performed an in-depth study of circulating and cutaneous immune cells by imaging mass cytometry, high-parameter flow cytometry, transcriptomics, and targeted serum proteomics. Low-dose IL-2 stimulated various circulating immune cells, including Treg cells with a skin-homing phenotype that appeared in the skin of SLE patients in close interaction with endothelial cells. Analysis of surface proteins and transcriptomes revealed different IL-2-driven Treg cell activation programs, including gut-homing CD38+, skin-homing HLA-DR+, and highly proliferative inflammation-homing CD38+ HLA-DR+ Treg cells. I'm thrilled to share our latest research published in the Turkish Journal of Biochemistry on the differential effects of choline on TLR2/4 mediated signaling through the regulation of Toll-interacting protein (TOLLIP) in hepatocellular carcinoma (HCC) cell lines. Our study with Ayşe Banu Demir, found that choline, known for its anti-inflammatory properties via the α7 nicotinic acetylcholine receptor (α7nAChR), exerts varying effects depending on the differentiation stage of HCC cells. Specifically, choline downregulated TOLLIP in both well-differentiated HEP3B and poorly differentiated SNU449 cells. It decreased the expression of α7nAChR, NF-κB, IL-6, TLR2, and TLR4 in well-differentiated cells, while increasing these markers in poorly differentiated cells. These findings suggest that choline could have therapeutic potential in the earlier stages of HCC by modulating TOLLIP and TLR2/4-dependent signaling pathways. This research highlights the complexity of choline's impact on HCC and opens new avenues for targeted therapies in liver cancer.

As Can Sucak Lab, we published the largest PID cohort ever studied in Turkiye. The study includes more than 300 patients, of those 57 novel, 127 disease-causing variants and more. A total of 44 Campylobacter isolates including 41 C. jejuni, two C. coli and one C. lari were used in this study. Although all of the C. jejuni isolates were verified by hipO based PCR assay, only 18 of the 41 C. jejuni were detected as positive by ceuE based PCR assay. Both of the methods showed negative reaction with C. coli and C. lari isolates. A new study recently published in Genome Medicine has unveiled significant genetic connections between Inflammatory Bowel Disease (IBD) and Parkinson’s Disease (PD). The research team, led by Meltem Ece Kars, Yuval Itan, and Inga Peter, employed advanced genomic analysis techniques to investigate rare genetic variants in patients diagnosed with both conditions. Their discovery of shared mutations in the #LRRK2 gene sets a new precedent for integrated treatment strategies. This research not only deepens our understanding of the genetic underpinnings common to these chronic diseases but also pioneers the development of dual-disease therapies. The FDA has approved fidanacogene elaparvovec-dzkt (#Beqvez™) as a one-time gene therapy to treat adult patients with moderate-to-severe hemophilia B, according to Pfizer. Eligible patients include those who: - Use factor IX (FIX) prophylaxis therapy, or - Have or have had life-threatening hemorrhage, or - Experience repeated, serious spontaneous bleeding episodes, and, - Do not have neutralizing antibodies to AAVRh74var, an adeno-associated virus (AAV), which can be screened via an FDA-approved test. “A one-time treatment with Beqvez has the potential to be transformative for appropriate patients by reducing both the medical and treatment burden over the long term,” said Adam D. Hello LinkedIn community, I am reaching out to seek guidance from experienced bioinformaticians regarding a couple of challenges I am facing in my cancer research project, I've been exploring several databases, including GEO, TCGA, and others, but have encountered some limitations. Here are the specific areas where I need assistance:: 1- Accessing mRNA Data for Prostate Cancer and Olaparib Treatment: I am in search of mRNA data from prostate tissues, both normal and tumor, before and after treatment with Olaparib. Despite extensive searches, I have encountered limitations in accessing such specific datasets. Could anyone advise on the best approach to locate or acquire this data even for paid ones? Any insights or resources would be immensely valuable. 2- General Strategies for Finding Cancer Transcriptomic Data: Additionally, I am looking for guidance on how to find transcriptomic data related to specific cancer treatments, such as mRNA data for breast cancer patients treated with Doxorubicin. I am open to exploring paid options if necessary, as this data is crucial for my ongoing project. Any advice on navigating these data limitations or suggestions for reliable databases would be greatly appreciated. Thank you in advance for your support and expertise!

Extremely happy to share that our recent work on " Early events during aggregation of Aβ16-22-derived switch-peptides tracked using Protein Charge Transfer Spectra," which has been published in the Elsevier journal "Analytica Chimica Acta" (I.F. 6.2) Here, we investigate the early events of Aβ aggregation using Aβ16-22 derived switch-peptides (SwPs) through a novel label-free approach employing Protein Charge Transfer Spectra (ProCharTS) in real-time. Excited to announce that our latest research article, under the leadership of Mehmet Ziya Doymaz and with contributions from Elif Karaaslan and Nesibe Selma Güler Çetin, has been accepted for publication in the prestigious Medical Microbiology and Immunology! 🎉 In this study, we investigated the cellular immune responses triggered by the nucleoproteins of the Hazara virus (HAZV) and its close relative, the Crimean-Congo Hemorrhagic Fever Virus (CCHFV). Our findings highlight significant parallels, along with some notable differences, in the immune response mechanisms between these two viruses. This further supports the use of HAZV as a model for studying CCHFV and contributes to the development of therapeutic strategies.
