Carcinoplacental alkaline phosphatase (CPALP), including three subtypes, such as Regan, Nagao and variant, has been found mainly by electrophoretic procedures. Using polyacrylamide gel slab of 5% (w/v) concentration, CPALP (Regan, Nagao) and placental alkaline phosphatase are electrophoresed next to the place of bone type isozyme or at the same place of it, and must needs to be differentiated by other methods, such as heat treatment.<br>Using polyacrylamide gel electrophoresis, CPALP was found as the fastest migrated isozyme band to the anode, when the polyacrylamide gel concentration of above than 8% is used. Thus, a polyacrylamide gradient gel electrophoresis method was introduced. Using this modified method of 16-8% gel concentration, CPALP could be detected as the fastest migrated isozyme band to the anode, and subbands of CPALP could also be identified.
