Your reading comprehension needs work. The diagnosis of clinical depression does not involve studying the chemicals in your brain. They don't even look at them. It involves studying your observable depressive traits such as feeling sad, having insomnia, fatigue, feeling worthless, not enjoying things, suicidal ideation, etc. You can see a list of them here. That you have a chemical imbalance is then simply assumed to be the cause.
