Suntan Drug Greenlighted for Trials
A drug that stimulates the body's tanning response — turning pasty skin caramel for up to two months — has been approved for human trials, but not for tanning.
Although the drug will not be available for cosmetic purposes any time soon, similar compounds are already being widely abused on the pharmaceutical black market. The official product, a man made hormone called afamelanotide, has received U.S. government approval to begin clinical trials.
"It's a bioabsorbable implant that you just inject into the skin," said Colin Mackie, director of business development for Clinuvel, the company bringing the drug to the U.S. "It stimulates melanin production."
Melanin is the body's natural pigment. It's responsible for the color of skin and protects humans from harmful solar radiation. The drug will be tested as a treatment for patients who face serious danger from the sun's rays, like those with rare genetic diseases or who must take immunosuppressants, Mackie said.
But online retailers already offer a similar (but not identical) compound under a more descriptive brand name, Melanotan II, which American and British health officials warn are already being abused.
"Using it could be dangerous to short and long-term health," British health officials said in November. It has not been tested for safety, quality or effectiveness and we don't know the potential side effects yet."
This week two British Medical Journal researchers warned that injecting the compound could change the size and shape of moles, which under normal circumstances can be a precursor to skin cancer.
Melanotan's plight highlights the difficulty in bringing drugs with large recreational or cosmetic potential to market for decidedly more medical conditions. Before the drugs can make it through safety and efficacy trials, rogue users — almost like pre-release movie filesharers — begin to test the drug on themselves. These high-demand, off-label uses could make regulatory agencies reluctant to approve the pharmaceutical for legitimate uses.
Both Melanotan I and II are synthetic cousins of a naturally occurring hormone that stimulates the production of melanin, the skin's pigment. But Melanotan II, in particular, appears to have aphrodisiac and erectile function effects as well.
First created and tested back in the late '80s at the University of Arizona, the synthetic tanning hormone received media attention from around the world. Tom Brokaw, in a 1991 broadcast, said that medical researchers had announced "what could be the answer" to tanning without the skin damage associated with the process. Wired first featured melanotan back in 2002, dubbing it the "Barbie drug" for its purported ability to induce weight loss and increase libido. The label has stuck and is often used in the press
But the drugs were a good idea in search of a disease. Fair-skinned people's desire to be tan is not a disease, and the tanning behaviors it induces could cause skin damage and cancer. Now, though, Clinuvel has found five, admittedly obscure, medical conditions that the drug could treat.
Over the last several years, the Clinuvel's CEO, Philippe Wolgen, has battled to make his drug sound less and less sexy.
"This will not be a cosmetic drug,” Wolgen told a biotech trade publication last year. “I oppose systemic treatment for cosmetics.”
He changed the company's name from the provocative Epitan to the boring Clinuvel. It might not make for great advertising copy, but the CEO's moves appear to have paid off.
"The FDA's decision is a landmark event in Clinuvel's growth," Wolgen said in a release this week. "Today's progress reflects some of the choices we had to make in our program early on in 2006 when changing the direction of the company."
But even as he tries to position his drug to be taken seriously, industry analysts following his company know that the drug could be a blockbusters on the cosmetics market.
After all, once a drug is approved by the FDA for one purpose, it's easier to extend it to new conditions, just like the glaucoma-treatment turned eyelash enhancer.
"[Melanotan] has potential as a 'Life Style' drug for those millions of people who prefer just a darker tint," a German analyst wrote back in 2004 [pdf]. "Market research has shown that the product has blockbuster potential in particular when used as a 'Life Style' drug, which draws a strong comparison to Botox or Viagra, two products with booming sales today."
Hopeful tanners took it upon themselves to deliver on that blockbuster potential as Melanotan languished in the pharmaceutical industry's basement.
Recent British media reports indicate widespread use of the compound. Purportedly, the drug is ordered online and comes in a white powder that is mixed with sterile water. That combination is injected into the abdomen.
One woman told the Daily Mail that she intended to keep using the injections despite their unknown provenance and possible side-effects.
"At first, injecting myself in my tummy seemed strange, but now I don’t even think about it — the needle is so fine I hardly feel it," she said. "And I am so pleased with the results that despite the adverse publicity I’m going to continue with them."
An online forum dedicated to Melanotan has more than 50,000 posts, most of which are about "Usage and Experimentation." Many describe detailed regimens for attaining darker skin and/or improved sexual function.
Here in the United States, Melanotan II appears widely available online, although the FDA warned Tennessee's Melanocorp, which bills its product as "100% US Made Melanotan II," to stop shipping to Americans in 2007. The company continues to ship to European countries.
The company provides a dozen or so purported before-and-after pictures, like the photo above. Though the results could be suspect, they look eerily similar to results obtained in trials at the University of Arizona or by Clinuvel (images at left). In early animal trials, injections of the drug caused a yellow dog's fur to become black and visibly darkened a frog's skin in minutes.

I have one of the "obscure" conditions that this drug might help. It's not all that rare (10-20% of northern hemisphere population may have some degree of it). I am happy this drug is being tested for a medical treatment instead of cosmetic uses.

This drug may actually have the effect of preventing some skin cancer. First assume that in trials it proves to be safe. A drug that gives a real tan for months would probably discourage sun tanning and tanning bed use and therefore reduce the number of people suffering sun damage to the skin.
I happen to be very fair skinned and burn easily, if my melanin could be increased in a safe way I would have that added protection on top of what I currently do to prevent sun burns. I'm already high risk for skin cancer, anything I can do to potentially decrease that risk would be helpful. Again this all assumes it proves reasonably safe in clinical trials, I would never take a gamble with a drug that hasn't been thoroughly tested (though that isn't a guarantee of complete safety just look at Vioxx but it's a better gamble than untested by far).

What happens to freckled people with this drug? With sun, my skin doesn't dark much, but my freckles darken and get bigger. It's a bit freakish.
By itself, more melatonin should reduce skin cancer since it is a natural sun blocker.

People who have fitzpatrick type I & II skin (those who can't tan at all and those who have difficulty tanning) are not simple able to go outside to get some color because all they do is burn!
When these sorts of folks use a melanotan peptide they are for once able to tan. This new ability for them allows them to spend time with their friends out in the sun, going to the beach, etc.
This is a big part of the reason why the melanotan peptides are such a big deal.

It's funny, you know seeing causcasian people scorn Michael Jackson for bleaching his skin, yet it is a national pastime for them to go and try to get tanned, what hypocrisy! Just like it is ok for people like Sarah Palin to hunt and kill moose and wolves as a sport but Michael Vick goes to prison for fighting dogs. The fact is, whatever excuse people use for this drug as well as the excuse they use for using tanning beds, know that "we" are paying attention to you and the way you treat people of color while at the same time you try your best to be like us, not the other way around.

Well, this is a great drug, but this is also what you get when the idiots at the FDA who have no ideas what they are doing and are in the pay of the pharmaceutical companies, get in the way of a legitimate product. This is the same type of crap they continue to pull with us parents with kids (taking away our cold and cough medicine) and those of us who use dietary supplements (specific examples include ephedra and l-tryptophan they banned). Give the FDA time and they'll take tylenol, motrin and cortisone cream off the market. The FDA should be disbanded and reformed with REAL consumers, not bureaucrats who take payment from the AMA and big Pharma.

Ugh..this is nothing new..I purchased "tanning pills" in 1988 through mail order out of a magazine and I had a fantastic bronze tan all summer and winter. The only problem is that when they start to wear off your skin gets a very unattractive mottled look..brown and whitish splotches..BUT if you dont mind looking like a faded Dalmation for a month then go for it..the end result really was great and no sun !!

wow, so it comes to this... as someone who once upon a time enjoyed getting a tan. stopped in my mid 20's because of all the talk of skin cancer. i opted for the self tanners and used as much sunscreen as possible for years. and then it happened. what i thought could never ever happen did. what you think happens to them not u happened to me. yes, skin cancer. thankfully it was caught in it's earliest stages and was removed. i can't begin to tell all of u how horrible it is to be diagnosed with any type of cancer. i thought i took pretty good care of my skin. my face especially. i had 5 stitches from the corner of my left eye straight across my nose. and the all i had was what appeared to be was a dot the size of pin.
i once upon a time felt the same way most of u do - who cares so long as it works and i look good attitude. i don't wish skin cancer or any type of cancer or any other illness on anyone - your all responsible for yourselves. but be warned - it can be, rest assured, be on of u next.
look good today, this summer, tomorrow next summer suffer the consequences.

With cancer rates steadily climbing we surely don't need to be putting more chemical compounds into our bodies without years of trials. There are products that have been on the market for ages until it's been realized they cause cancer; then they are removed, too late for some.

I feel there is nothing wrong with this drug. If it is used for medical breakthoughs or vanity, who cares?! Some people that have commented on not looking "third world" have issues with other races. There are thousend of people wo test this drug and there was never a serious health problem.

Some people do not no what it is like to have very light skin. I live south of Houston Texas and in the summer I can only spend 30 min outside in the sun before I start burning. Normal day of outside activity take two coats of sunscreen through the day. Me tan is like most people in the dead of winter. It would be nice to have a little added protection from the sun. Spending time in the sun to get tan is just dumb, cancer, cancer, cancer. Up north with higher declination of the sun I'm just fine. But i like to do water sports all year round. So here i live were only darker people ought to.

Hey, here's a real medical issue question... Does Clinuval do anything with Vitamen D? Because of all the sunscreen we are using, many of us are seeing lower Vitamen D production... which happens when the skin is exposed to UV rays. It would be a positive side effect IF the use of this drug stimulated Vitamen D production in a tail-wagging-the-dog sort of way.

I have been around the world of dermatology my entire life and I can not believe that our government would allow something that could obviously stimulate skin cancer and who knows that else to become much greater and/or worse than by not using this "hot" product. I would never buy this drug and would never recommend it to anyone. I think this might be just as ridiculous as rubbing butter onto a burn instead of ice. wtf.

You are absolutely correct Greg! Light skin does help you look less third worldish as you like put it. However this the natural taning of sking comes from years of working outside, in the sun. Building things, working the fields, herding, creating new jobs not by sitting infront of a computer trying to find the quickest way to make money and how to work less and send the economy down the drain, while not setting a single foot out the door. On the drugs use I am completely for the use of it towards medical solutions and if it is safe enough to avoid leasure exposure to the sun I am for it too. I know lots of ppl who tan and none of them have come even close to appearing any other race. Especially since overtanning produces more of a yellowish orange tint, definitely not black. Really?!?! I live in Miami and the sun is constantly shining obviously and I'll tell you something, true Floridians do not go to the beach as much as people think. Tanning is dangerous (and I mean people who lay out for days and days and days), we are not in the 60's anymore people, that much sun exposure to the skin is not good especially for people with fair complexions. If you have a disease where your doctor prescribes this drug I guess I can understand but American's are already abusing prescription drugs and I believe this will be added to that list. Being in the sun and fresh air for a little while is healthy for you and if you go for a run or walk around the block a few times a week during the day over time your skin will build its own immunity and gradually darken. I've used Melanotan II. I got it literally one week before it's sale was made illegal in the United States.
It works. It works, VERY WELL. The only problem was the 'viagra' side effect. I had to take it at night so that I wasn't out in public afterwards. Other than that it's been over a year since I used it and guess what, I'm still alive, nothing is wrong. I'd rather something help my cells produce melanin on their own than lie out in the sun or a tanning bed which requires twice as much effort, less quality results and makes me way more susceptible to skin cancer than this stuff.
Do they even have one instance of this drug having harmful side effects on humans at all? No, only on animals. And believe me, thousands of people have used this I've been to the forums. The FDA can suck it.

i have a skin condition that makes my skin on my hands and feet purple, it really helps when i tan for the color to change back to a normal skin tone, but tanning isn't healthy to do too frequently. it would be amazing to get a couple shots and not have to worry about people staring at my hands and feet anymore. i hope that after clinical trials, that it is approved by the FDA and that it becomes available. i would pay a lot of money to not have to deal with the embarassment anymore...

Yes, it's funny, I remember growing up I was ashamed of my white skin, I would pour tanning oil all over my body in the shower, then years later I didn't want to be tan anymore, so I stopped going out in the sun, I'd work out indoors and I'd run on the tredmil, but then I got white again and didn't like it so I started eating a lot of carrots, which work better than this drug and aren't dangerous.

I'm Indian (Trinidadian Indian) and my skin is brown, my hair is black, my nose is unfortunately a little too big for my face, but that's what I was born with and I wouldn't change it. Humans have yet to be able decide what color or not they are going to be before they are born. Now granted, two people of a different race/color can mate, but that doesn't always guarantee what color or not the kid is going to be; so my advice is to shake your fist at God, maybe he will light your butts up with electricity and change your colors forever.

I'm siding with J on this one. I also have vitiligo and it has been diagnosed as "profound, incurable" as it covers more than 30% of my body. My doctors and I have tinkered around with many different medications over the years to try and trigger my body into producing more melanin. After several failures, I opted towards a more pale complexion to reduce it's appearance. But I would still be willing to try a new product. Many of the products on the market were designed for the treatment of diseases and have been misused for vanity, but this does not negate it's true intentions and the benefits for people like me. Hmmm, maybe I can get my friends in Germany to send me some. I'm game, bring it on!

mmmkish, I don't really think it's about criticizing people for changing. I believe that people are too heavily involved in wanting to change that they will go to unhealthy lengths to do so. I'm recieving a masters in toxicology and injecting your body with any drug in general is dangerous in some way. It can be prescribed to you but it's not natural to your body. Even the food we eat is full of chemicals these days. Adding more unnecessary materials to your body is not healthy. However you are right, noone should critcize anyone who is that unhappy with something about themselves. As long as they do not hurt themselves trying to change it.

I read this article because i thought it was interesting as it has always been psychologically speaking to see the way in which an attraction to something like color in skin usually live in conjuction with a dissatisfaction in something color in skin. I think that is where all the black folks feel like..."what the frick." Historically and presently we have felt the sting of the projected dislike of our natural beauty and many for generations will try to undue that internalization that has been passed forward as well as through our life experiences. It just seems like shedding light on it is good but not in an evil way, since evil never solves anything. But I understand, people are angry when they feel they have been wronged. I would just like to say that all of the 'minority' people that are calling white people hypocrites for wanting to be tan are retarded. When we are putting on sunless tanner or going tanning in a bed most of us are not trying to look black or hispanic. Has anyone thought, though as sick as it is that this racial discussion is happenning because maybe it's just time. I mean we need to be a little more understanding about it all but maybe people are just tired of not talking about it like we've been doing for decades. Vitiligo is an absence of melanocytes in the skin. This hormone stimulates melanocytes. Most likely, it would not change a thing for people like Michael Jackson. Unless, there are actually dormant melanocytes that could be stimulated.

Granted, the dangers of the hormone are, so far, unknown. Studies could reveal those risks to be safer than being under tanning lights or in the sun. Its not hard to see that people, who burn easily,(like me) could reap benefits from not burning every summer, too. That is why it is in the study phase. To find out if it is a safer alternative to the more dangerous tanning methods,that we have right now.

In a free country, the citizens make their own decisions without meddling by everyone else! This drug could prove to be preventative for skin cancers! Then, everyone and his brother could use it! But even if approved by the FDA, it still, like all medicines, could carry some risk! It is matter as to how much benefit one gets for the risk.(risk/benefit ratio)
My wife is one of those people that wants to have a tan (she says tanned fat looks better than white fat") And she is an absolute doll. Thing is, as long as it is a free country, its no body else's business how she gets tanned. Especially, if there is a safer way to do it!

This low-level "discussion" of race baiting has not lived up to the scientific discourse that "Wired Science" encourages. For those of you who hate whites, I say, LIGHTEN UP! For those of you who hate people of color (especially blacks) I say, TONE IT DOWN! I'm talking about your hateful rhetoric, not your skin color. Words matter, because they are a direct reflection of your personal psychological (some would say "spiritual") state. Hate has been proven scientifically as well as accepted theologically to be self-destructive, so GET OVER YOURSELVES! You don't help other's spiritual/psychological state, either. Check in with your Maker, if you believe, before you key out this B.S. over the internet.
Furthermore-- I think most intelligent people would not even bother to post any comments of a scientific or sociological nature here, because of the cretins who have clogged up these comments (for the most part) with B.S. Kudos to those of you who have at least pleaded for tolerence here.
I'm just as guilty in a way, though. My first knee jerk reaction was to think about how the US and other NATO military establishment types could exploit this stuff for the "benefit" our fair-skinned soldiers. Maybe they could freeze and cut off their moles first, (to keep them from becoming carcinogenic) and then ORDER them to take regular injections for high altitude, full of UV rays, sunny Afghanistan and Iraq, to mitigate skin cancer later-- and to blend in with the indigenous population if necessary, for our special forces on certain missions in Pakistan where they can successfully reach out and "touch" al-Qaida. Heck, it might even work for CIA types in the Khyber Pass area, too! If the military could bypass the slow FDA approval, then they could follow up with studies through the V.A. hospitals later, to see if skin cancer incidents are above the statistical standard deviation for the general popluation from which these soldiers and agents came, or from the rest of the military personnel with which they were serving. Such an epidemiological study could serve to confirm or refute any FDA approval or rejection as a follow-up study. Remember-- one key to scientific inquiry is to confirm or refute established hypotheses with additional scientific analysis. This sounds "cold", but much of medical science is cold as well-- and so is war.
As for the FDA, it will be interesting to see how powerful the sun tan lobby might be, when (not if) their "scientific" studies funded by them confirm that melantonin II is a carcinogenic agent, while the pro-melatoninII lobby funds their studies to scientically "prove" that the stuff is not carcinogenic. This way, they can sell it for legitimate medicinal uses first, and for the big buck "lifestyle" reasons later. No wonder the undermanned and underfunded FDA has to take so long to get any new drug approved or disapproved! They have to rely on "privately funded" studies by lobbies like these most of the time, and seldom get to do their own objective research on drugs "up" for human trials.
As for comments about President Obama: The major reason that he is our President, regardless of whether you agree with his policies or not, is because the man is supremely "confident in his own skin", and he shows it. This is why he was elected by us "unwashed masses" in my humble opinon. I may have some concerns about the wisdom of some of his policies, but it cannot be denied that he has long since put stupid racial issues far behind him, and has moved on. He is simply TOO BUSY for racial hatred, and TOO INTELLIGENT to waste any more time on it than he has to give the issue of bigotry. At first analyis, this guy is a PRAGMATIST, not a hater.
Shame on you bigots for turning this into a stupid discussion about your own insecurities, projected on people not like you. Shame on the rest of us for allowing it to happen-- and not commenting intelligently on the scientific and sociological ramifications of this development as the Wired Science blog aspires us to do.
Next time, let's all try to do a little better. Thanks for your attention to my own rants.

personally, I'm holding out for a robot body that I can download my consciousness into and then change into any color or form, but thanks for that skin-tanny drug anyway, Science.
I know. Crazy huh. But ask yourself whats worse: Being a robot in a humans' world or being a human in a world where people get into arguments over skin color whilst discussing a drug that gives you a tan?

I'm not a fan of cosmetic pills or surgery. If they want to test this drug for real illnesses, I'm completely for it. But I wouldn't use it. I like my pale skin.
Also, I'm appalled at all the "racism" that everyone seems to think is associated with a tanning pill. A white person wanting to be tan does not mean they want to be black.

This article makes some great points about the need for proper trials. So many drugs show promise early on, yet fall at one hurdle or another as they are tested, and tested again, for safety and effectiveness.
It would be a mistake to think that these peptides are necessarily the finished product. If you're thinking of trading in your sunbathing or sunbeds for an injection of an untested biological agent, please think again - you're just jumping from the frying pan into the fire. Stick to the fake tan if you *must* look brown.

I love this stuff my buddy from the uk sends it to me every now and then, no more uv fake tanning no more getting burnt outside and it looks as if you just came back from vacation tan.

@Nina, I have not problem with dark skinned people. I was making a snide remark about a system that is discriminating against light skinned individuals and is not in fact alleviating the suffering of the black poor, but rather just benefiting the black elite.
